We know that protein is a macromolecule in living organisms, it is the material basis of all life, and plays a very important role in cell structure, function, metabolism, model conduction, etc., in the field of biochemistry, the study of protein function and structure is an important topic.

Protein extraction is the first step in the study of protein. In the extraction process, to overcome the cell wall obstacles, destroy the cell membrane to extract protein from the cell. The extraction experiment has a direct impact on the follow-up protein detection and purification experiment, so the successful extraction of high-quality protein is the success foundation of full experiment.
Centrifugation is very important during the extraction process
We always got some questions from user that in protein extraction experiments, can I avoid centrifuge? The answer is No. Because there will be some insoluble substances such as cell debris after cell lysis, and centrifugation must be carried out to separate cell debris and organelles, etc., and the extracted protein may be impure if it is not centrifuged.

Centrifugation is very important during the extraction process
Why use a high-speed refrigerated centrifuge?
Centrifugation generates a certain amount of thermal energy, and protein may be denatured and enzymatic hydrolysis in a hot environment, so a high-speed refrigerated centrifuge is required. Low temperature conditions help prevent protein denaturation, improve the accuracy of separation and purification, and preserve their native structure and function. Operating at low temperatures helps reduce protein degradation and inactivation, ensuring that biological activity remains in subsequent applications.

Then why high speed is necessary?
For large molecular weight proteins, low-speed centrifugation, such as the extraction of nuclear proteins, can avoid damage to protein. For example, when performing WB centrifugal protein test, the speed range of the centrifuge is usually 1000~20000RPM, especially for the extraction of membrane proteins, a higher centrifugation speed is required to ensure the full precipitation of proteins.
In the process of protein purification, high-speed refrigerated centrifuges are indispensable equipment.
Below we share some steps on how to extract protein from mouse intestinal tissue.

●Prepare an appropriate protein extraction lysate according to the sample volume and store it on ice.
●Fresh tissue: Use clean scissors after sterilization to chop the tissue block as much as possible, cut it small, put it into a homogenizer, add it to the lysate prepared in advance, fully homogenize and lysate, and then put it on ice.
●Lyse for 30 minutes, mix every 10 minutes.
●Precool high-speed refrigerated centrifuge at 4 °C, set speed at 12000 rpm at 4 °C, and set time of 15 minutes for centrifugation.
●After centrifugation, take the supernatant and transfer it to another micro tube, do not suck the sediment, the supernatant is the protein solution.

It is common to use centrifuge in most biological experiment, it is necessary to choose the right instrument.
